Introduction: The protein CD86 (Cluster of Differentiation 86) is a molecule expressed on antigen-presenting cells that provide costimulatory signals necessary for T cell activation and survival. It is the ligand for two proteins at the cell surface of T cells, CD28 antigen and CTLA-4 (cytotoxic T-lymphocyte-associated protein 4). CD86 is also known as B7.2. Its principal mode of action is by binding to CD28. Along with CD80, these molecules provide the necessary stimuli to prime T cells against antigens presented by antigen-presenting cells. Binding of this protein with CTLA-4 negatively regulates T-cell activation and diminishes the immune response.
Principle of the Assay: The microtiter plate provided in this kit has been pre-coated with an antibody specific to sCD86. Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody preparation specific for sCD86 and Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. Then a TMB (3,3\',5,5\' tetramethyl-benzidine) substrate solution is added to each well. Only those wells that contain sCD86, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm +/- 2 nm. The concentration of sCD86 in the samples is then determined by comparing the O.D. of the samples to the standard curve.
