Intended Uses: Testosterone ELISA is an enzyme immunoassay for the quantitative measurement of Testosterone in serum and plasma (EDTA. lithium heparin or citrate plasma).
Principle of the Assay: Testosterone ELISA Kit is a solid phase enzyme-linked immunosorbent assay (ELISA) based on the principle of competitive binding. The microtiter wells are coated with a monoclonal (mouse) antibody directed towards an unique antigenic site of the testosterone molecule. During the first incubation. the testosterone in the added sample competes with the added enzyme conjugate. which is testosterone conjugated to horseradish peroxidase, for binding to the coated antibody. After a washing step to remove all unbound substances. the solid phase is incubated with the substrate solution. The colorimetric reaction is stopped by addition of stop solution, and optical density (00) of the resulting yellow product is measured. The intensity of color is inversely proportional to the concentration ofthe analyte in the sample. A standard curve is constructed by plotting 00 values against concentrations of standards, and concentrations of unknown samples are determined using this standard curve.
Samples: Serum and plasma
Assay Type: Quantitative Competitive
Detection Range: 0.083-16 ng/ml
Sensitivity: 0.083ng/mL. Testosterone ELISA or ELISA (enzyme-linked immunosorbant assays) Kits in general, are a valuable research tool for a myriad of applications in a range of scientific settings. Currently, three major types of ELISA formats are used by researchers: sandwich, competitive and indirect. Most commercially available ELISA Kits are sandwich or competitive. Commercially available ELISA Kits contain wells that have been pre-coated with the capture antibody. Please refer to the product manual for the ELISA format of your specific kit.
