The ORF73/HHV8 n/a (Catalog #MBS200144) is an Antibody produced from Mouse and is intended for research purposes only. The product is available for immediate purchase. The ORF73/HHV8 antibody reacts with Human and may cross-react with other species as described in the data sheet. MyBioSource\'s ORF73/HHV8 can be used in a range of immunoassay formats including, but not limited to, ELISA (EIA), Western Blot (WB), Immunohistochemistry (IHC). Researchers should empirically determine the suitability of the ORF73/HHV8 n/a for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. The ORF73/HHV8 n/a product has the following accession number(s) (GI #2246532) (NCBI Accession #AAB62657) (Uniprot Accession #O40947). Researchers may be interested in using Bioinformatics databases such as those available at The National Center for Biotechnology Information (NCBI) website for more information about accession numbers and the proteins they represent. Even researchers unfamiliar with bioinformatics databases will find the NCBI databases to be quite user friendly and useful.

Please refer to the product datasheet for known applications of a given antibody. We\'ve tested the ORF73/HHV8 antibody with the following immunoassay(s):
Western Blot (WB) (Transfected and non-transfected 293T cell lysates were resolved by SDS-PAGE, transferred to NC membrane and probed with anti- ORF73/HHV8 (1:1,000) and anti-MBP (1:1,000). Proteins were visualized using a goat anti-mouse secondary antibody conjugated to HRP and an ECL detection system.)
Immunohistochemistry (IHC) (Paraffin embedded sections of Kaposi\'s sarcoma tissue were incubated with anti-ORF73/HHV8 (1:50) for 2 hours at room temperature. Antigen retrieval was performed in 0.1M sodium citrate buffer and detected using Diaminobenzidine (DAB))
Western Blot (WB) (The cell lysates were resolved by SDS-PAGE, transferred to PVDF membrane and probed with anti-human ORF73/HHV8 antibody (1:1000). Proteins were visualized using a goat anti-mouse secondary antibody conjugated to HRP and an ECL detection system.
Lane 1.: 293T cell lysate (40ug)
Lane 2.: ORF73/HHV8-MBP tag Transfected 293T cell lysate (5ug))
Western Blot (WB) (The cell lysates were resolved by SDS-PAGE, transferred to PVDF membrane and probed with anti-human MBP antibody (1:1000). Proteins were visualized using a goat anti-mouse secondary antibody conjugated to HRP and an ECL detection system.
Lane 1.: 293T cell lysate (40ug)
Lane 2.: ORF73/HHV8-MBP tag Transfected 293T cell lysate (5ug))
ORF73 protein of human herpesvirus 8 (HHV-8) is a kaposi\'s sarcoma-associated herpesvirus (kSHV) latent protein which has been suspected of playing a crucial role in modulating viral and cellular gene expression. ORF73 is associated with cellular chromatin and stays on the chromosomes during cell division. It maintains the viral genomes during cell division by tethering the viral episomes to the chromosomes. It binds directly to replication origin recognition complexes (ORCs) that are primarily associated with the terminal repeat (TR) region of the HHV-8 genome.
Antigen Species: Human
Immunogen: Recombinant human ORF73/HHV8 (122-329aa) purified from E. coli. Clone Note: Anti-human ORF73/HHV8 mAb, clone AT4C11, is derived from hybridization of mouse F0 myeloma cells with spleen cells from BALB/c mice immunized with a recombinant human ORF73/HHV8 protein.
Usage: The antibody has been tested by ELISA, Western blot and IHC analysis to assure specificity and reactivity. Since application varies, however, each investigation should be titrated by the reagent to obtain optimal results.
