The LCK lck (Catalog #MBS603187) is an Antibody produced from Mouse and is intended for research purposes only. The product is available for immediate purchase. The Lck, p561ck (Lymphocyte-specific Tyrosine Kinase) reacts with Human, Mouse, Rat and may cross-react with other species as described in the data sheet. MyBioSource\'s Lck, p561ck can be used in a range of immunoassay formats including, but not limited to, ELISA (EL/EIA), Western Blot (WB), Immunoprecipitation (IP).Suitable for use in Western Blotting, Immunoprecipitation, Kinase Assays. Antibody preparations from which the BSA has been omitted have been used successfully as a coating antibody in ELISA.,
Dilution: ELISA: 1:100-1:1000
Western Blotting: 1.0ug/ml with an incubation of 2 hours at RT
Immunoprecipitation: 2.0 ug/mg of protein lysate. Researchers should empirically determine the suitability of the LCK lck for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process.
The LCK lck product has the following accession number(s) (GI #349702) (Uniprot Accession #P06239). Researchers may be interested in using Bioinformatics databases such as those available at The National Center for Biotechnology Information (NCBI) website for more information about accession numbers and the proteins they represent. Even researchers unfamiliar with bioinformatics databases will find the NCBI databases to be quite user friendly and useful.

p56lck is myristoylated at its N-terminus, then translocated to the plasma membrane where it is palmitoylated and targeted to lipid rafts. p56lck associates with several proteins including c-Cbl through its SH3 domain and the cell surface glycoproteins CD4 and CD8 through its N-terminal domain. p56lck is expressed predominantly in T-cells and its function is critical both for T-cell development in the thymus and activation of mature T-cells in the periphery by antigen. The activity of p56lck is regulated by phosphorylation of two conserved tyrosine residues, tyrosine 505 (equivalent to tyrosine 529 of c-Src) and tyrosine 394 (equivalent to tyrosine 418 of c-Src). p56lck activation can increase responsiveness of some T cell hybridomas to antigen. The phosphorylation status and the activity of p56lck is regulated by the tyrosine protein phosphatase CD45, a protein which is also known as the leukocyte common antigen (LCA). The gene for p56lck is located in a chromosomal region which is susceptible to breakage and translocation. Abnormalities in this chromosomal region are often observed in lymphomas and neuroblastomas.
Immunogen: Recombinant protein corresponding to amino acid residues 1-255 of murine p56lck protein. Hybridoma: Produced by fusion between BALB/c mouse splenocytes and mouse myeloma p3-NS1-Ag4-1 cells. In general, we may offer more than one antibody to a given target to enable options for the researcher. Available antibodies recognizing LCK are readily searchable from our website. Different antibodies against the same target such as LCK may be optimized or tested for different applications and species. This enables researchers to select the option that may be best for their model system, to screen more than antibody to determine which one may be best for their model system, as well as to use more than one antibody to follow up on and validate their results.
