Flow Cytometry: 0.25ug labels/10e6 cells. Tested in MCF-7 cells.
Western Blot: 2ug/ml detects human FOLR1 in human cortex tissue and HeLa cells under non-reducing conditions only.
Immunocytochemistry: 8-25ug/ml. Tested on immersion fixed MCF-7 cells.

Lysates were prepared from Hela cells in non-reducing sample buffer, resolved by SDS-PAGE (30ug total protein/lane), and transferred to an Immobilon-P membrane. The blot was developed with MBS605669 (2ug/ml) and chemiluminescent detection substrate.)
Testing Data (Detection of FOLR1 in MCF-7 human cell line. FOLR1 was detected in immersion fixed MCF-7 human breast cancer cell line using MBS605669 (10ug/ml) for 3 hours at RT. Cells were stained with the DayLights 557-conjugated Anti-Mouse IgG Secondary Antibody (red) and|counterstained with DAPI (blue).)

FOLR1 is a 37-42kD protein that mediates the cellular uptake of folic acid and reduced folates. Dietary folates are required for many key metabolic processes including nucleotide and methionine synthesis, the interconversion of glycine and serine and histidine breakdown. Mature FOLR1 is an N-glycosylated protein that is anchored to the cell surface by a GPI linkage. Human FOLR1 shares 83% amino acid sequence identity with mouse and rat FOLR1. FOLR1 is predominantly expressed on epithelial cells and is dramatically up- regulated on many carcinomas. It is critically required during early embryogenesis as shown in knockout mice which die in utero with gross morphological defects. FOLR1 is internalized to the endosomal system where it dissociates from its ligand before recycling to the cell surface. A soluble form of FOLR1 can be proteolytically shed from the cell surface into the serum and breast milk.
