All antibodies recognize D-dimer in ELISA. All MAbs recognize D-dimer in Western blotting under non-reducing conditions. MAbs DD22, DD41, DD44, DD46 and DD189 interact with beta-chain of D-dimer in Western blotting under reducing conditions. MAbs DD93 and DD255 interact with gamma-chain of D-dimer in Western blotting under reducing conditions.
Recommended pairs to be used in a sandwich immunoassay for D-dimer detection in human plasma (capture-detection):
One-step sandwich immunoassays:
DD189 - DD255 (Equal specificity to D-dimer and high MW fibrin degradation products). DD2 - DD41 (Slightly more specific to high MW fibrin degradation products at high concentrations).
Two-step sandwich immunoassays*:
DD2 - DD4 (Approximately equal specificity to D-dimer and high MW fibrin degradation products).
(*To be analyzed in a two-step sandwich immunoassay, plasma must be diluted 1:1 or more with 10 mM Tris-HCl buffer, pH 7.5, containing 1 M NaCl and 0.1 % Tween 20. To avoid non-specific binding the final NaCl concentration in plasma samples must be 0.5 M or more.). Researchers should empirically determine the suitability of the D-Dimer n/a for an application not listed in the data sheet.
