The CDC45 cdc45 (Catalog #MBS619873) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The Cdc45 reacts with Human, Monkey, Mouse, Rat and may cross-react with other species as described in the data sheet. MyBioSource\'s Cdc45 can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB).Suitable for use in Western Blot.
Dilution: Western Blot: 1:1000. Incubate membrane with diluted antibody in 5% w/v BSA, 1X TBS, 0.1%
Tween-20 at 4 degree C with gentle shaking, overnight. Researchers should empirically determine the suitability of the CDC45 cdc45 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process.
The CDC45 cdc45 product has the following accession number(s) (GI #4502713) (NCBI Accession #NP_003495.1) (Uniprot Accession #O75419). Researchers may be interested in using Bioinformatics databases such as those available at The National Center for Biotechnology Information (NCBI) website for more information about accession numbers and the proteins they represent. Even researchers unfamiliar with bioinformatics databases will find the NCBI databases to be quite user friendly and useful.

Western Blot (WB) (Western blot analysis of extracts from various cell types using MBS619873.)
The initiation of DNA replication in mammalian cells is a highly coordinated process that ensures duplication of the genome only once per cell division cycle. Origins of replication are dispersed throughout the genome and their activities are regulated via the sequential binding of pre-replication and replication factors. The origin recognition complex (ORC) is thought to bind to chromatin throughout the cell cycle (1,2). The pre-replication complex (Pre-RC) forms in late mitosis/early G1 phase with the binding of CDT1 and cdc6 to the origin, which allows binding of the heterohexameric MCM2-7 complex. The MCM complex is thought to be the replicative helicase and formation of the pre-RC is referred to as chromatin licensing. Subsequent initiation of DNA replication requires the activation of the S-phase promoting kinases cdk2 and cdc7. Cdc7 phosphorylates MCM proteins bound to chromatin and, in conjunction with CDT1, recruits the replication factor cdc45 (3-5). Cdc45 is required for formation of pre-initiation com- plexes at the G1/S transition and for activation of replication origins. The level of cdc45 protein expression is associated with the proliferative status of the cell or tissue. Terminally differentiated and senescent cells lack cdc45 and highly proliferative cell lines express high levels of cdc45 (6).
Immunogen: Synthetic peptide corresponding to the carboxy terminus of human cdc45 protein (KLH-coupled). In general, we may offer more than one antibody to a given target to enable options for the researcher. Available antibodies recognizing CDC45 are readily searchable from our website. Different antibodies against the same target such as CDC45 may be optimized or tested for different applications and species. This enables researchers to select the option that may be best for their model system, to screen more than antibody to determine which one may be best for their model system, as well as to use more than one antibody to follow up on and validate their results.
