The Apaf-1 apaf1 (Catalog #MBS440041) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The Apaf-1 (A15) Antibody reacts with Human and may cross-react with other species as described in the data sheet. MyBioSource\'s Apaf-1 can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB). Researchers should empirically determine the suitability of the Apaf-1 apaf1 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. The Apaf-1 apaf1 product has the following accession number(s) (GI #20141188) (NCBI Accession #O14727.2) (Uniprot Accession #O14727). Researchers may be interested in using Bioinformatics databases such as those available at The National Center for Biotechnology Information (NCBI) website for more information about accession numbers and the proteins they represent. Even researchers unfamiliar with bioinformatics databases will find the NCBI databases to be quite user friendly and useful.

Large multi-protein complexes known as the DISC (death inducing signalling complex) and the apoptosome activate the normally inactive caspases through proteolytic processing (1). The formation of these complexes leads to the activation of caspases and the execution of apoptosis (1,2). The apoptosome is a large 700 or 1400 kDa protein complex comprised of Apaf-1 (apoptotic protease activating factor-1), cytochrome c, and dATP (1). Apaf-1 and cytochrome c in the presence of dATP leads to the oligomerization of Apaf-1 and the formation of either the 700 or 1400 kDa complexes (3&4). Once formed the apoptosome activates caspase-9, which in turn activates the effector caspases (1&5). Some researchers have found the 700 kDa apoptosome to be the most effective activator of the effector caspases, when compared to the 1400 kDa apoptosome (3&4). Apaf-1 has also been shown to be a target of regulation by a variety of proteins that include p53, IAPs, heat shock proteins and Smac/Diablo (1&6).
Origin: Apaf-1 (A15) is provided as an affinity purified rabbit polyclonal antibody, raised against a peptide mapping to the amino terminus of human Apaf-1. Immunogen: Synthetic peptide mapping at the amino terminal domain of human PCNA. In general, we may offer more than one antibody to a given target to enable options for the researcher. Available antibodies recognizing Apaf-1 are readily searchable from our website. Different antibodies against the same target such as Apaf-1 may be optimized or tested for different applications and species. This enables researchers to select the option that may be best for their model system, to screen more than antibody to determine which one may be best for their model system, as well as to use more than one antibody to follow up on and validate their results. Breast Neoplasms, Colonic Neoplasms, Disease Models, Animal, Inflammation, Liver Diseases, Liver Neoplasms, Necrosis, Neoplasms, Experimental, Nerve Degeneration, Nervous System Diseases are some of the diseases may be linked to Apaf-1 (A15) Antibody. The following patways have been known to be associated with this gene.
