Dilution: Immunohistochemistry: 1:100-1:500; Antiserum was used on perfusion fixed tissue. Perfusion: 1) Calcium-free Tyrode\'s solution, 2) paraformaldehyde-picric acid fixative, and 3) 10% sucrose in PBS as a cryo-protectant. Desired tissues were dissected and stored overnight in 10% sucrose in PBS.
Slide mounted tissue sections were processed for indirect immunoflourescence. Slides were incubated with blocking buffer for 1 hour at room temperature. Primary antiserum was diluted with blocking buffer to the appropriate working concentration. Blocking buffer was removed and slides were incubated for 18-24 hours at 4 degree C with primary antiserum. Slides were rinsed 3 times and then incubated with secondary antibodies for 1 hour at room temperature. Slides were again rinsed 3 times and cover-slipped. Staining was examined using fluorescence microscopy. Researchers should empirically determine the suitability of the ADRA2A adra2a for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process.

Alpha 2A, 2B, and 2C adrenergic receptors are seven transmembrane receptors that couple to inhibitory G-proteins. Alpha 2C has a wide distribution pattern in the CNS. On neurons alpha 2 adrenergic receptors are located on both pre- and postsynaptic domains where they mediate an inhibitory role in the central and peripheral nervous system. Alpha 2C has a wide distribution pattern in the CNS. At the spinal cord level the vast majority of alpha 2C is not of primary afferent origin; however, a small fraction of neurons in DRG do express mRNA for alpha 2C and harbor the protein. Colocalization has been observed between alpha 2C and enkephalin in the spinal cord dorsal horn, suggesting that alpha 2C may be expressed by a subset of spinal interneurons. The distribution pattern of alpha 2C suggests functions at several levels in the brain. Antibody showed positive staining in rat spinal cord dorsal horn, ventral horn motor neurons and retina. When examined in other brain regions known to express alpha 2A staining was not observed.
