AEC is one of the most important chromogenic reagents for peroxidase. The coloration which is showed to be fresh red, and applies to immunohistochemistry and various dot detection. Easy to solution in organic impregnant is its defect, so, we recommend users to seal slide with water soluble sealing reagent in case of using AEC.
Assay Procedure: 1. After the peroxidase detection, wash with TBS or PBS 4 times and each time for 5 minutes. 2. Add one drop of chromogenic reagent An and B respectively into 1ml of distilled water and mix evenly Add it to the sample and colorate in dark for 10-30 minutes. If no background appearing, keep coloration until satisfied. 3. Wash fully with distilled water to stop reaction. Counterstain with methyl blue for 3-5 minutes or nonalcoholic hematoxylin (Mayer\'s) if necessary. 4. Seal sllide with water soluble sealing reagent (MyBioSource can provide), or glycerol. Observe with microscope.
Product Note: Counterstain with alcoholic hematoxylin is not recommended.
