Typical Testing Data/Standard Curve (for reference only)
Principle of the assay: AATF ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-AATF antibody and an AATF-HRP conjugate. The assay sample and buffer are incubated together with AATF-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the AATF concentration since AATF from samples and AATF-HRP conjugate compete for the anti-AATF antibody binding site. Since the number of sites is limited, as more sites are occupied by AATF from the sample, fewer sites are left to bind AATF-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The AATF concentration in each sample is interpolated from this standard curve.
Samples: Serum, plasma, Cell Culture Supernatants, body fluid and tissue homogenate
Assay Type: Competitive
Detection Range: 2.5-50ng/mL
Sensitivity: 0.1ng/mL. Intended Uses: This AATF ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Guinea pig AATF. This ELISA kit for research use only, not for therapeutic applications!. Guinea pig Apoptosis Antagonizing Transcription Factor ELISA Kit or ELISA (enzyme-linked immunosorbant assays) Kits in general, are a valuable research tool for a myriad of applications in a range of scientific settings. Currently, three major types of ELISA formats are used by researchers: sandwich, competitive and indirect. Most commercially available ELISA Kits are sandwich or competitive. Commercially available ELISA Kits contain wells that have been pre-coated with the capture antibody. Please refer to the product manual for the ELISA format of your specific kit.
